Résumé :
Identifying functional protein sites and understanding which and how genetic variations alter them are central questions for bioengineering, medicine, and fundamental biology. Protein sequences, 3D shapes and motions have been exploited for decades to address these questions. I will present several computational methods to identify protein functional sites, in particular interacting surface regions, and to predict mutational outcomes, potentially at very large scale. I will discuss the relative contributions of sequence- and structure-based information in addressing these problems.
